I think Raine and and Reina are beautiful. I know a preschooler with the middle name Raine, and I think it's lovely. To me, though, Reina and Raine are COMPLETELY different names. As you said, one means queen, and one is a nature name. They're in totally different categories in my head--even the sound of them.

I prefer Raina, nickname Rain. Rain is pretty and all, but I can't imagine trying to be an adult with it, it doesn't feel complete.

I like Rain fine. I know a little girl named Rainey, and that's pretty cute, but not sure about an adult named Rainey? I guess it works.

I think if you're going to go with Rain, you should spell it Rain. It's pronounced the same either way (Rain/Raine/Rayne etc), but no matter what the name is, I think it's always classier to spell it correctly/the original way.

I love the simplicity and imagery of Rain. IMO it looks like you're trying too hard if you add any other letters to it.

I prefer Reina (though I associate it with being a Japanese name, wasn't aware it was also Spanish). If you're going to go with Rain though, I would just spell it Rain. Raine just seems more complicated and I've only seen it as a boy's name (ie, Raine Wilson on "The Office").
